Zymo Research Corporation Quick-DNA Plant/Seed 96 Kit (2 x 96 Preps)
The procedure is easy and can be completed in as little as 50 minutes: plant samples ( 80 mg each) are added directly to the tubes of a ZR BashingBead™ Lysis Rack (2.0 mm) and are rapidly and efficiently lysed by bead beating (e.g., 2010 GenoGrinder Instrument, page 5) without the use of organic denaturants or proteinases. Polysaccharides and polyphenols/tannins are removed from the DNA using our Zymo-Spin™ 96-well plate and Silicon-A™-HRC Plate technologies, respectively. The eluted DNA is ideal for downstream molecular-based applications including PCR, arrays, etc.

There is no need for glass beads, phenol, or vortexing. Instead, plasmid DNA is reliably recovered from yeast cells whether colonies, patches on plates, or liquid cultures are sampled. Plasmid yields are typically between 0.01-0.3 ng for most 2 based plasmids from 1.5 ml overnight cultures. This kit also works well with low copy number yeast plasmids.
